Citric Acid Determination

چکیده

Three types of methods have been proposed for the determination of citric acid in urine. Salant and Wise (1) used the unspecific mercuric sulfate reagent of Deniges (2) in a study of the toxicity of citrates. Thunberg (3) developed an enzymatic method employing a dehydrogenase said to be specific for citrate. The remaining methods depend on the so called pentabromoacetone method of Stahre (4), a qualitative test published in 1897 which Kunz (5), in 1914, used as the basis of the first quantitative pentabromoacetone method. It has been generally overlooked that in 1847 Cahours (6) prepared a bromination product from several citrates which he called “bromoxaforme” which according to his melting point and solubility data is without doubt identical with pentabromoacetone. Cahours was aware of the specificity and analytical applicability of the reaction. (“Le brome peut done servir a reconnaftre de petites quantit& d’acide citrique mhlangbes a l’acide tartrique.“) Kunz’s technique was adapted for use with urine by Amberg and McClure (7). Fasold (8) with this method was the first to isolate citric acid from normal urine. Kometiani’s (9) modification of this method was applied to urine by Stillmann and Schaerer (10). Pucher, Sherman, and Vickery (11) developed a spectrophotometric method based on the color formed when pentabromoacetone is treated with sodium sulfide. Our procedure is another modification of this method. It is precise, accurate, and timesaving. The “pentabromoacetone reaction” has been clarified and new reagents with certain advantages have been introduced. While the procedure was designed for 15 cc. samples of urine, it is also applicable to 2 cc. samples, as well as to other fluids of comparable citrate content. The range is from 1 to 40 mg., although with care, useful results are obtainable down to 0.2 mg. The method has already been used in a study of the variation of urinary citrate with the menstrual cycle (12).

Simultaneous kinetic spectrophotometric determination of citric and ascorbic acid by H-point standard addition method

The H-point standard addition method was applied to kinetic data for simultaneous determination of citric and ascorbic acid or selective determination of ascorbic acid in presence of citric acid. The method is based on the difference in the rate of reaction of citric and ascorbic acid with copper(II)-ammonia complex. The linear dynamic ranges for the two analytes of citric and ascorbic acid are...

A colorimetric method for the determination of citric acid in blood and plasma.

As early as 1897 the oxidative bromination of citric acid to form a brominated acetone derivative, presumably principally pentabromoacetone, was adapted to the quantitative determination of citrate (1). Most subsequent procedures have been based on the same reaction. Among the more recently described procedures are those of Pucher (2) and of Goldberg and Bernheim (3). These methods, with minimu...

Development of Better-Quality Assay Method for the Citric Acid and Sodium Citrate in Ophthalmic/Oral Solutions and Their Application to Deformulation Studies

There were several techniques determined for the analysis of citrate and citric acid mixtures in the pharmaceutical dosage forms. Titration methods, photometric and ion chromatographic methods were used for their determination. These methods will restrict too many factors where the accurate quantification of citrate and citric acid is extremely challenging. Citric acid is the natural flavor use...

مقایسه اسیدهای آلی با وزن مولکولی کم در توصیف آزاد شدن روی در تعدادی از خاک‌های آلوده

The rate of metal transfer from the solid phase to solution is an important factor governing their concentration in the soil solution and its availability. In this research, the release rate of Zn in contaminated soils from Isfahan was studied using solutions citric acid, oxalic acid and malic acid 0/01 M during the period of 2 - 504 hours and its relationship with soil characteristics was inve...

Micro-determination of isocitric and cis-aconitic acids in biological material.

oxaloacetate +pyruvate ci 8-acontate = ocitrat. In order to be able to examine the new hypothesis we have worked out methods for the micro-determination of ci8-aoonitic and i8ocitric acids, a preliminary account ofwhich has been published in the Proceedings of the Biochemical Society (Krebs & Eggleston, 1943 b). So far no specific micro-methods for the estimation of the two. acids have been des...

Chromatographic Determination of the Acids of the Citric Acid Cycle in Tissues* by Charles

Many methods are to be found in the literature for the determination of several of the individual acids of the “citric acid cycle;” however, to date no method has been available for the simultaneous determination of all of the acids in a single sample of tissue. Isherwood (1) employed partition chromatography on silica gel to separate and determine subsequently a group of acids present in fruit...
